For DC, I love Green Lantern, Green Arrow, Supergirl, Superboy, and the Justice Society. I'm also huge on the 90's characters they created. Good on them for at least trying, and a fee of them were pretty cool. You had Sovereign Seven, the Ray, Damage, Loose Cannon, and in my opinion the best of the batch: Aztek. Aztek is great if you've never read it. It was short but good. Co-written by Grant Morrison and Mark Millar.
For Marvel, I'm all about my green-skinned lady: She-Hulk. I also dig almost all of them so I'll just say X-men to keep it simple (but Cable, Bishop, and Beast are top of that list). Machine Man, 90's Ghost Rider, 90's Punisher as well as Max Comics and the current Edmondson volume, and Iron Man in the 70's and 80's. Spiderman pre-Clone saga.
I tend to be era or writer specific. Larry Hama Wolverine was cool, but Tieri wasn't. Modern Spidey doesn't float my boat but a lot of the 70's, definitely 80's, and some of the 90's stuff is wonderful. Batman hits full stride from the early 1980's to about 2004.

Marvel: Carnage, Venom, The Blob. I always thought that Carnage and Venom were awesome villains. And the Blob well I like him for more personal reasons.
DC: Joker, Batman, Scarecrow. Batman is an obvious fan favorite including my favorite hero. Scarexrow is down right cool in all aspects especially since the beginning of the Arkham games. Joker is my favorite villain ever personality and lines wise he is a brilliant character.

Marvel would have to Spider-Man always been a fan since I was a little kid and for a villian the Green Goblin pretty basic and for DC I really am a fan of Batman and my favorite villian from there is The Joker and TBH I also like Scarecrow and Mad Hatter. Edit: had to pick Batman and the Joker as favorites becuase who can beat their chemistry Joker with obsession with Batman trying his hardest to manipulate him and Batman never really letting the Joker mess with his mind and just kicks the crap out of him but never try's to kill him.

OP didn't specify it had to be a Marvel or DC character so... (EDIT: actually he did in the subject line...oh well...too late to take my post back) Butcher from The Boys Wee Hughie from The Boys Glen from Walking Dead The Punisher (but only if it is the Garth Ennis version) Spider from Transmetropolitan Ennis' disdain for the superhero dominated market is what made Punisher and The Boy's so good. When Ennis left Punisher Max and Swierzynski took over, the character fell back on a slightly more heroic leaning where Ennis portrayed Frank as a less-bad bad guy. Swierzynski is fine at spinning crime yarns, but his also felt too generic and he was afraid of the hyperbolic highs that Ennis hit. The Barracuda miniseries was my favorite Punisher Max story for that very reason. Sillier like his Marvel Knights run but with f*ck words galore. I would say that the Jason Aaron Punisher Max is a worthwhile read. It isn't Aaron's best work (that'd be Scalped) but it gets the Ennis tone more closely than anyone who has followed. I'm digging Edmondson's current run which wrapped this week. If they want Punisher to be a hero, they have to humanize him. And they did. It was an approach that worked for a more empathetic human story.
